ELISA: BENCHMARK IN SERODIAGNOSIS
  • ELISA (Enzyme-linked immuno-sorbent assay) is a type of immunoassay, used for detection of Antibodies, Antigens , Proteins ,Peptides , Biomolecules
ELISA !!!! Why called so?-
  • Antigen  or antibody of interest is absorbed on to plastic surface (‘sorbent’)
  • Antigen is recognised by specific antibody  or vice versa (‘immuno’)
  • Antibody molecule is attached with an enzyme (‘enzyme-linked’)
  • Substrate added at the end of the reaction
  • Enzyme breaks it to coloured end products
Generations of ELISA
  • 1st generation - Infected cell lysate
  • 2nd generation - Glycopeptides (recombinant antigen)
  • 3rd generation  - Synthetic peptides
  • 4th generation – Both Antigen and antibody detected
Applications
  • Diagnosis of infections – almost any
  • Detecting allergens in food and house dust
  • Detection of autoantibodies  - collagen vascular diseases
  • Measuring toxins in contaminated food
  • Detecting illicit drugs like Cocaine , opiates or anabolic steroids
CONCLUSION:
  • Rapid diagnostic assays have a lot of drawbacks
  • Need of the hour : SHIFT FROM RAPID DIAGNOSTIC TESTS TO ELISA
  • Only drawback of ELISA is turnaround time
